ABSTRACT

Objective To design a kind of suitable equipment to collect the specimens from endoscopic mucosal resection (EMR),and observe the effect of the collector on clinical application. Methods The six-hole rotating collector was made from transparent glass,in which the specimens could be aspirated through the channel and remained in a mental net. A total of 813 cases with 1254 lesions in esophagus,stomach and large intestine were included in the study. All the cases were male 516,female 297,age in 7-90 years old (average 53.2 years). The collector was used for receiving these EMR specimens that less than 1cm in diameter. The final diagnoses were proved by histopathologic examination. Results The rotating collector was named as Transparent 6-hole Rotating Eodoscopic Specimen Collector,and registered as national patent. The collector was a round column type,about 160g in weight and 56 mm?60 mm in size,which was light,clear and easily cleaned,and worked well during clinical application. The total collecting success rate for specimen receiving is 93.3%. All the specimen samples kept undamaged and good for histological analysis. Conclusion The rotating specimen collector is useful and economic for endoscopic mucosal resection.

ABSTRACT

Malignant transformation is closely related to gene mutations.The tumor suppressor gene p53 codes for a transcription factor,P53 protein,plays a critical regulation role in oncogene expression,DNA synthesis and repair systems,and cell apoptosis.It has been demonstrated that p53 mutation occurs in more than 50% of cancer cells,which is thought to play a crucial role in malignant transformation.p53-based gene therapeutical approaches in vitro and in vivo aiming at a direct correction of the specific molecular defect in p53-mutated malignant cells,showed notable efficacy in numerous cancer types.In this review,we discussed the current advances in this field,focusing on adenovirus-mediated wild-type p53 gene therapy.

ABSTRACT

Tumor angiogenesis, resulting from the imbalance of angiogenic cytokines and angiogenesis inhibitors, is crucial for tumor growth and metastasis. As a new field of cancer therapy, tumor anti-angiogenesis therapy with regard to abrogation of tumor vasculature has become an important anti-tumor strategy. Gene therapies have the potential to provide antiangiogenic mediators specifically targeted to organs containing tumors and make a great improvement in this therapy. This article reviews the current understanding of tumor angiogenesis, with a focus on gene therapy in this field.

ABSTRACT

Portal hypertensive gastropathy (PHG), a term used to describe the endoscopic appearance of gastric mucosa with a characteristic mosaic-like pattern, is characterized entities that can be associated with gastrointestinal blood loss in patients with portal hypertension. More than 65% of patients with portal hypertension from cirrhosis will develop PHG,however,it could also occur in the setting of non-cirrhotic portal hypertension. In patients with portal hypertension, the incidence of PHG was associated with severity of liver disease and the presence of both oesophageal and gastric varices. The exact etiology of PHG is not clearly defined, the diagnosis of PHG depends on endoscopy and histology, therapy of PHG is directed at lowering portal pressure by ?-blockers or shunt procedures.

ABSTRACT

Objective: Acute pancreatitis(AP) often impairs peripancreatic tissues and involves more distant organs,such as the liver,which would exacerbate the progression of the disease.This study aimed to explore the clinical characteristics of acute pancreatitis-associated liver injury.Methods: We randomly retrieved 156 cases of AP from the medical records retrieval system of Jinling Hospital,of which 102 were complicated by liver injury.The latter were divided into a severe AP(n=44) and a mild AP group(n=58) for comparative analysis of the severity of liver injury and time of recovery.Results: Liver injury occurred in 65.38% of the AP patients,and it was more serious,had a longer disease course and took more time to recover in the severe than in the mild AP group.Conclusion: The incidence and severity of acute pancreatitis-associated liver injury are positively correlated with the severity of acute pancreatitis.

Objective:To explore the expression of Ezrin and CD44v6 in gastric carcinoma and their correlations to clinicopathologic features. Methods:The expression of Ezrin and CD44v6 were detected by immunohistochemistry in gastric carcinoma(n=53) and normal gastric mucosa(n=20).Results:①The positive expression rates of Ezrin in gastric carcinoma and normal gastric mucosa were 56.6% and 30.0%.While the positive expression rates of CD44v6 in gastric carcinoma was 58.5% and no expression in normal gastric mucosa.②The expression of Ezrin was correlated to the differentiation(P0.05).③The expression of Ezrin was positively correlated with that of CD44v6 in gastric carcinoma(rs=0.740,P

ABSTRACT

The Wnt signaling path has a function of delivering the growth incitement signal,and it is closely related with the growth and differentiation of cells.The unusual activation of the Wnt signaling path may cause abnormal propagation and differentiation of cells and consequently the development of tumor.Besides,this signaling path serves an interactive function in different human tumors.Understanding the contribution of this signaling path in tumor formation may afford a basis for the clinical diagnosis and early intervention of tumor.

ABSTRACT

Objective: To evaluate the contents of ? aminobutyric acid(GABA) and glutamate(Glu) in gastric cancer tissue and normal tissue surrounding the cancer. Methods: 30 patients of gastric cancer underwent surgical treatment were examined. A high performance liquid chromatographic method(HPLC) was applied for the analysis and quantification of GABA and Glu in gastric cancer tissues and normal tissues surrounding the lesion after precolumn derivatization with phenylisothyocyanate (PITC). GABA and Glu were determined with UV detector. The differences in GABA and Glu contents in gastric cancer tissues were regarded to have close link of lymph node metastasis and histological type. Results: GABA and Glu contents were high significantly in gastric cancer tissue comparing with those of surrounding normal tissue( P 0.05). Conclusion: GABA and Glu are involved in the factors of gastric carcinogenesis. The results suggest GABA Glu system detection should be included into the diagnosis and therapy consideration in gastric cancer.

ABSTRACT

Objevtives: To explore the relationship between ? aminobutyric acid (GABA) and the proliferation or differentiation of jejunal epithelial cells. Methods: Immunohistochemical expression of GABA, glutamate decarboxylase (GAD, including two isoforms, GAD65 and GAD67) was investigated in rat jejunum. Meanwhile, double staining was performed with GAD65 immunohistochemistry followed by lectin histochemistry of fluorescent wheat germ agglutinin. Furthermore, cell kinetics was evaluated by immunohistochemistry using a monoclonal antibody to proliferating cell nuclear antigen (PCNA). Results: Immunoreactive GABA and GAD65 were distributed in the villi of jejunum, while GAD67 was negative in epithelial cells. Strong positive staining of GABA and GAD65 was mainly localized along the brush border of jejunal villi. In addition, there existed a few GABA and GAD65 strong positive cells scattering in upper portion of the jejunal villi. Double staining showed that immunoreactive GAD65 was not found in goblet cells. PCNA staining nucleoli were found in the lower and the middle portions of the jejunal crypts. Therefore, GABA and GAD65 were expressed in maturation or functional zone. Conclusion: It suggests that GABA might be involved in the regulation of differentiation and maturation of epithelial cells in rat jejunum.

aminobutyric acid(GABA) is a principal inhibitory neurotransmitter in the mammalian central nervous system(CNS). In recent years,some studies have demonstrated that GABA is also present in various peripheral tissues,including the gastrointestinal tract. GABA acts as a gut hormone in addition to its role as an enteric neurotransmitter,which involves in the transport and regulation of intracellular information in stomach. Therefore,it inhabits development and growth of gastric cancer. Glutamic acid decarboxylase(GAD) is a major GABA synthesizing enzyme. This review will narrate the relationship between GABA,GAD and gastric cancer.

Somatostatin,a cyclopeptide hormone,is generally distributed in vivo.Some studies have indicated that somatostatin can reduce proinflammatory cytokine in hepatic stellate cells and extracellular matrix,and inhibit their proliferation and contraction,promote their apoptosis.It can also regulate the function of Kupffer cells,influence the homing of hepatic stem cells.Somatostain has therapeutic effect on liver fibrosis,especially on the liver fibrosis caused by schistosoma mansoni.

ABSTRACT

Objective To evaluate the effect of Caspase-1 inhibitor on hepatic tissue and on the expression of hepatic interleukin-18 of rats with experimental severe acute pancreatitis(SAP).Methods Forty-two SD rats were randomly assigned into 3 groups: healthy control group(HC,n=6),SAP+normal saline group(SAP-S,n=18) and SAP+ICE inhibitor group(SAP-ICE-I,n=18).SAP was induced by retrograde infusion of 4% sodium taurocholate into the pancreaticobliliary duct in SD rats.Animals in HC group received similar surgical procedure and duct cannulation but without sodium taurocholate inoculation.In SAP-S group,rats received the first intraperitoneal injection of isotonic saline 2 hours after induction of acute pancreatitis,and the same injection was repeated at 12th hour.ICE inhibitor was injected into the rats in SAP-ICE-I group 2 hours after induction of acute pancreatitis,and the same injection was repeated at 12th hour.Blood samples were obtained from the rats in SAP-S and SAP-ICE-I groups via cardiac puncture at 6,12 and 18h after modeling,respectively.The levels of serum amylase(AMY),alanine aminotransferase(ALT) and aspartate aminotransferase(AST),and the peritoneal fluid,were detected at these three time points.The histopathology of pancrease and liver were observed under light microscope.Intrahepatic expression and localization of IL-18 protein were detected respectively by Western blotting and immunohistochemistry.Results Serum AMY,ALT,AST and peritoneal fluid were significantly increased in SAP-S and SAP-ICE-I groups compared with those in HC group(P

ABSTRACT

Objective To explore the role of Kupffer cell-expressed Fas ligand(FasL) on acute pancreatitis(AP),and the protective effect of gadolinium chloride(GdCl3),a Kupffer cell inhibitor,on liver injury during AP.Methods Fifty-four ICR mice were randomly assigned into three groups: healthy control group(n=6);AP group(four time points for observation,6 mice each);Gd+AP group(GdCl3 pretreatment group,four time points for observation,6 mice each).AP was induced in mice using cerulein.Liver parenchymal enzymes(AST,ALT and LDH) and amylase(AMY) were assayed using an automatic analyzer.Serum FasL was assayed with ELISA and FasL protein expression in liver was assayed by Western blotting.Results After induction of AP in mice,the serum levels of AST,ALT,LDH,AMY and liver FasL expression were significantly elevated.Serum FasL levels at 4,8,16 and 24h after AP induction were 505.94?36.21,496.60?33.65,476.64?22.66 and 450.75?38.21,respectively,which were significantly higher than those in healthy control group(83.60?7.75,P
